摘要: 替代脂肪源的开发和利用是解决鱼油短缺问题的必然选择。然而,随着替代水平的提高,鱼体常常表现免疫水平和抗病能力降低。鱼油替代的本质为脂肪酸替代,深入研究脂肪酸与鱼类免疫的关系显得尤为重要。本实验综述了脂肪酸对鱼类免疫性能的影响及调控机制。饱和脂肪酸会降低鱼类免疫力,而适量添加 n-3 长链多不饱和脂肪酸( LC-PUFA) 、共轭亚油酸( CLA) 或提高 n-3 /n-6 多不饱和脂肪酸( PUFA) 的比例有利于鱼体免疫力发挥; 饲料中脂肪酸主要通过细胞膜结构、信号传导、类花生四烯酸、细胞因子和类固醇激素等途径对鱼类免疫进行调控。脂肪酸与鱼类的免疫性能具有高度相关性,而调控机制的研究尚有较大空间。未来研究应该侧重于以下几个方面: 脂肪酸对免疫相关转录因子的调控机制; 鱼类肠道脂肪酸组成改变与菌群结构和免疫性能之间的相关性; 环境因子对鱼体脂肪代谢和免疫力的影响; 非脂肪酸成分( 矿物质、维生素) 对鱼类脂肪酸代谢和免疫过程的调控机制。

当前水产动物病害严重,提高或保持水产动物自身免疫力是抗病的基础,而免疫力与营养和饲料有关[1],尤其脂肪酸是营养元素中影响免疫的重要因子,因此脂肪酸与免疫的关系成为新的研究方向。尤其在当前鱼油替代普遍,新脂肪源大量涌现的情况下,越来越多的证据表明,鱼油被新脂肪源替代后会影响鱼类免疫活力[2 - 4],这使得在当前深入研究脂肪酸与鱼类免疫的关系显得尤为重要。
1 脂肪酸种类和来源
脂肪酸是脂肪的重要组成部分,它是由一条长的烃链( 尾) 和一个末端羧基( 头) 组成的羧酸。烃链 不 含 双 键 ( 三 键 ) 的 称 作 饱 和 脂 肪 酸( saturated fatty acid,SFA) ; 具有 2 个及以上双键的称 作 多 不 饱 和 脂 肪 酸 ( polyunsaturated fattyacid,PUFA ) ; 具有 4 个及以上双键且碳链长度大于 20 的称作长链多不饱和脂肪酸 ( long chainpolyunsaturated fatty acid,LC -PUFA ) 。不同脂肪酸之间的主要区别在于烃链的长度( 碳原子数目) 、双键数目和位置。根据第一个双键距离烃链尾的碳原子数目可以将 PUFA 分为 n-3 系列和n-6 系列。 其中 n-3PUFA 主要包括亚麻酸 ( α-linolenic acid, ALA ) 、 二 十 碳 五 烯 酸( eicosapentaenoic acid,EPA) 和二十二碳六烯酸( docosahexaenoic acid,DHA ) ; n-6PUFA 主要包括亚 油 酸 ( linoleic acid,LA ) 和 二 十 碳 四 烯 酸( arachidonic acid,AA) 。水产饲料常用脂肪源大致分为鱼油、植物油和动物性脂肪,具体分类和脂肪酸组成详见表 1。
2 鱼类免疫系统
鱼 类 免 疫 系 统 包 括 固 有 免 疫 ( innateimmune) 和适应性免疫( adaptive immune) 系统,其中 固 有 免 疫 又 称 为 非 特 异 性 免 疫 ( non-specificimmune) ,是鱼类免疫的第一道防线[6],包括细胞免疫和非特异性体液免疫。免疫细胞又称白细胞,包括粒细胞、单核细胞和淋巴细胞 3 类细胞。其中粒细胞又可分为噬酸、碱、中性 3 种,其中中性粒细胞是血液中主要的吞噬细胞,细胞中含有大量的溶酶体酶。固有免疫指标主要包括吞噬指数、呼吸爆发、凝集素、溶菌酶、抗蛋白酶、C-反应蛋白、干扰素-1、转移因子以及补体,其中补体又包括替代途径补体和经典途径补体[7]。研究表明,鱼类的替代途径补体活力水平远远高于哺乳类[8],这说明鱼类比高等动物更依赖于替代途径补体来抵御病害入侵。
适应性免疫又称为特异性体液免疫,是鱼类受到抗原刺激后机体进行免疫应答产生特异性抗体的过程。在硬骨鱼中已发现鱼类具有完善的适应性免疫参数如 MHC、TcR 和 Ig 等,但某些鱼类由于对抗原刺激反应较低,不能产生抗体或产生量很少。对虹鳟( Oncorhynchus mykiss) 的二次攻毒实验表明,首次攻毒时鱼类主要以固有免疫进行防御,二次攻毒时鱼类的适应性免疫才发挥显著作用,这说明鱼类的适应性免疫需要较长时间的适应才能产生,具有明显的滞后性,因此鱼类主要利用固有免疫来发挥抗病作用[9]。
3 脂肪酸对鱼类免疫的影响研究进展
3. 1 多不饱和脂肪酸对鱼类免疫的影响
脂肪酸对鱼类免疫力的影响,已有了一些报道[4,10 - 11]。鱼类免疫力会受到饲料脂肪酸种类、剂量和比例的影响。一般来说,饱和脂肪酸会抑制鱼类免疫[10]; 饲料中添加适量多不饱和脂肪酸尤其是 n-3PUFA 时,鱼类的生长及免疫力均得到显著提高[3,12 - 13],但是当超过需求量时,鱼类的免疫力并不能进一步提高,而是出现免疫抑制现象[10],这可能是由于过量的 n-3LC-PUFA 降低了鱼类免疫器官表面病原识别受体( TLR22) 及其接头蛋白( My D88) 的表达所致[12],另一方面,过量的 n-3LC-PUFA 容易导致该种脂肪酸发生过氧化反应,超氧阴离子会攻击免疫细胞膜,从而大大降低其免疫性能[14]。Wang 等[11]发现,饲料中添加适量的维生素 E 能够显著提高牙鲆( Paralichthysolivaceus) 的免疫性能。
3. 2 功能性脂肪酸对鱼类免疫的影响
共轭亚油酸( conjugated linoleic acid,CLA)是亚油酸的一组含有共轭二烯键的空间和几何异构体,在鱼类中是被研究最多的功能性脂肪酸。根据双键的位置和空间构型的不同可将 CLA 分为多种异构体,其中顺 9,反 11 ( c-9,t-11) 和反10,顺 12( t-10,c-12) 是 2 种主要的异构体并已被证明具 有 生 物 学 活 性。在 人 类 和 哺 乳 动 物 中CLA 已被证实具有较为明显的抗炎、抗氧化和抗癌症作用[15-17]。而在水产动物上有关该脂肪酸对免疫性能的影响仅在少数几种鱼类,如舌齿鲈( Dicentrarchus labrax)[17]、大黄鱼 ( Larimichthyscrocea)[18]和草鱼 ( Ctenopharyngodon idella)[19]有所涉及,结果表明,适量 CLA 能够明显增强动物的免疫力。有关 CLA 各种异构体功能的研究在鱼类更为缺乏,哺乳动物中的研究证实顺 9,反11( c-9,t-11) 和反 10,顺 12 ( t-10,c-12 ) 这 2 种异构体具有不同的生物活性[20]。
3. 3 n-6 与 n-3 脂肪酸的含量与比例对鱼类免疫的影响
由于植物油中含有大量的 n-6 系列多不饱和脂肪酸,而相对缺乏 n-3 系列多不饱和脂肪酸,所以用植物油取代鱼油会导致 n-6 /n-3 比例发生改变,而高比例的 n-6 /n-3 会降低鲑( Salmo salar)的免疫力[3,21],这可能是由于高含量的 n-6 系列脂肪酸会增加鱼体炎症因子( IL-1β 和 TNFα) 的表达所致[3]。体内过度炎症反应会导致机体出现氧化应激,因此寻找适当方法调控炎症反应至适当水平成为治疗养殖鱼类“亚健康”亟待解决的问题。已有研究表明,上述由亚油酸引发的高炎症 水 平 可 以 通 过 添 加 部 分 共 轭 亚 油 酸( CLA)[18]或者适当提高饲料 ALA /LA 的比例来适当抑制[22]。Gjen 等[23]认为鲑中不同比例的n-6 / n-3 对抗病力没有显著影响,只是在鱼油和豆油等比例混合组( n-6 /n-3 约为 1. 6) ,鱼体会产生较高水平的 PGE2和 LTB4。造成这一差异的原因可能是不同的饲料基础配方、实验条件和实验鱼种类。Wu 等[24]发现,当饲料中 ALA 和 LA 总和 在 2% 且 二 者 比 例 为 3 时 点 带 石 斑 鱼( Epinephelus malabaricus) 的免疫性能最高。
适宜的DHA /EPA /AA 对于鱼类免疫也具有重要 意 义。鱼 类 饲 料 n-3 LC-PUFA ( DHA 和EPA ) 含量较丰富,AA 则相对缺乏,Harel 等[25]研究表明,饲料中缺乏 AA 会降低仔稚鱼的抗应激能力,从 而 抑 制 免 疫 系 统 发 育。因 此,在 富 含EPA 和 DHA 的饲料中,AA 会提高鱼的健康状况和质量,尤其是在高密度养殖中[26 - 27]。AA 发挥生理作用主要是通过 AA 的活性代谢产物类花生四烯酸如 PGE2和 LTB4来执行的。一定浓度的PGE2和 LTB4具有刺激免疫细胞增殖的作用,但是高浓度的 PGE2会对免疫产生抑制作用,而Koven 等[28]认为 AA 抑制免疫是由于体内代谢产生了高浓度的可的松,可的松作为一种肾上腺糖皮质激素,可以通过减少循环淋巴细胞的实际噬菌率来抑制免疫。此外,AA 还可以作为第二信使刺激巨噬细胞进行呼吸爆发。由于淡水鱼和海水鱼具有不同种类的必需脂肪酸以及不同的脂肪酸代谢途径,所以 DHA /EPA /AA 比例的确定在 2 种鱼中也不完全一样。在淡水鱼中,这一比例较难确定,由于淡水鱼类能够利用 ALA 合成EPA 和 DHA ,LA 合 成 AA ,因 此 确 定 其 适 宜DHA / EPA / AA 比例较为困难,且意义也不如在海水 鱼类 中 重 要。而 海 水 鱼由 于 不 能自 身 由ALA 合成 EPA 和 DHA ,LA 合成 AA ,因此 DHA /EPA / AA 比例的确定所受的干扰相对较小[29 - 30]。目前已得到一些海水仔稚鱼的最适 DHA /EPA 需求,然而该比例在海水幼鱼或早期成鱼阶段研究较少,有关该比例对机体免疫力和脂肪代谢的影响及 调 控 机 制 的 探 讨 更 是 少 之 又 少[31]。当DHA / EPA 在 1. 53 ~ 2. 08 时,花鲈 ( Lateolabraxjaponicus) 的生长、免疫和抗干露能力最强[32]。
4 脂肪酸的免疫调控机制研究进展
4. 1 细胞膜结构调控途径
细胞膜的结构骨架是磷脂双分子层,脂肪酸是磷脂的重要组分,因此,细胞膜的流动性取决于膜的脂肪酸组成[23]。细胞培养液中和食物中的脂肪酸组成会改变免疫细胞细胞膜的脂肪酸组成,进而影响鱼类免疫。Montero 等[2]发现鱼类吞噬活性与 n-3PUFA 成正比,与单不饱和脂肪酸( 主要是油酸) 和亚麻酸的量成反比。培养液中的 n-3PUFA 能够显著增加诸如淋巴细胞和免疫细胞等的流动性,而棕榈酸对 Caco-2 细胞膜的流动性却没有显著影响[33]。巨噬细胞脂肪酸组成与其在食物中的含量也呈剂量反应,鱼吞噬细胞中 AA 与食物中 AA 的比率维持在 3 ~ 7,而吞噬细胞中 EPA /食物 EPA 维持在 1 左右,而 DHA 的这一比例也维持在 2 左右。用不同的植物油取代60% 或 80% 的鱼油后进行摄食生长实验,结果发现添加豆油组实验鱼的巨噬细胞中亚油酸的含量有所提高,而添加亚麻酸实验鱼的巨噬细胞中亚麻酸含量也相应增加,替代组巨噬细胞中油酸的含量显著提高,而鱼油组巨噬细胞具有更高含量的 n-3PUFA。相应地,巨噬细胞的免疫功能如吞噬活性和呼吸爆发在鱼油组要显著好于植物油替代组,这说明巨噬细胞的脂肪酸组成与其免疫功能之间具有显著相关性[2]。
4. 2 信号转导途径
膜连接酶、受体或离子通道功能与细胞信号转导密切相关,脂肪酸可以作为第二信使或孔道、膜连接酶的调节亚基来影响免疫细胞的功能。在多核粒细胞的培养液中外源性添加花生四烯酸( AA) 能够刺激吞噬细胞和粒细胞进行呼吸爆发,这一方面是由于 AA 产生的生理活性物质类花生四烯酸能够调控免疫反应,另一方面 AA本身或者含有 AA 的磷脂酸以及二酯酰甘油可作为第二信使直接调节机体免疫[34]。细胞膜闭合蛋白能形成和维持细胞的稳固结合,EPA 能上调细胞膜闭合蛋白的产生,降低细胞对大分子的渗透,而 AA 和油酸则下调细胞膜闭合蛋白的表达。此外,膜连接酶的活性对脂肪酸环境特别敏感,如内皮细胞膜连接酶 Na,K-ATPase 对血管的收缩功能具有重要的作用,而 n-3PUFA 可抑制它的活性,而其他类型的脂肪酸作用较弱; 在 n-3PUFA中,DHA 对膜流动性和钠离子通道阻滞的作用较ALA 强[35]。
Toll 样受体( TLRs) 是存在于动物免疫细胞表面的一类病原识别受体,能够特异性识别病原关 联 分 子 结 构 ( pathogen-associated molecularpattern,PAM P) 如脂多糖 ( LPS ) 、肽聚糖、非甲基化 Cp G DNA[36]。当感受外界病原刺激时,TLRs可以将信号通过依赖 My D88 或不依赖 My D88 的途径进行信号传递,最终激活免疫反应,从而消灭外界病原[37]。已有研究表明,饲料脂肪酸水平或组成会影响 TLR22 和 My D88 的 mRNA 表达量来调 控 大 黄 鱼 免 疫 力 和 抗 刺 激 隐 核 虫 的 能力[12,31]。此外,脂肪酸也可以通过与 TLRs 的直接接触来调控该信号途径,且不同类型脂肪酸对TLRs 的激活作用不同,从而导致免疫调控能力的差异[38]。
4. 3 类花生四烯酸途径
类花生四烯酸是机体内部由 C20PUFA 衍生成的具有生理活性作用的物质,主要有前列腺素( prostaglandin,PG) 、凝血恶烷( thromboxane,TX)和白三烯( Leukotriene,LT) 。鱼体的磷脂酰肌醇( PI) 能够选择性积累 C20PUFA 如 AA 、EPA 和DHGLA ,故认为 PI 是类花生四烯酸前体物质的主要来源。磷脂酰肌醇的 sn-2 位上常常是 PUFA如 EPA 和 AA,在磷脂酶-2 ( PLA-2) 的作用下,AA 或 EPA 水 解 下 来, 在 环 氧 化 酶( cyclooxygenase, COX ) 和 脂 氧 合 酶( lipoxygenase,LOX) 的作用下 AA 生成前列腺素E2( PGE2) 和白三烯 B4( LTB4) ,而 EPA 生成前列腺素 E3( PGE3) 和白三烯 B5( LTB5) 。PGE2和LTB4的生物学活性明显高于 PGE3和 LTB5,具有提高免疫活性的作用。PGE2能够调节免疫细胞的功能,并且具有剂量效应。有研究表明,低密度的 PGE2( < 10- 9mol / L ) 对于维持正常的免疫功能和 T 细胞分化来说都是必需的; 而浓度超过10- 8mol / L 则会引起免疫抑制。LTB4在免疫调节中同样具有重要作用,LTB4是免疫细胞功能的有效调控者,能够促使 T 细胞和 B 细胞增殖,促使单核细胞和 T 细胞释放细胞因子如白细胞介素-1,此外还可以作为化学引诱物提高自然杀伤细胞的活性。同样,LTB4在低浓度( < 10- 9mol /L ) 时具有免疫调节作用,当浓度超过 10- 7mol / L时,则具有免疫抑制作用[39]。
免疫细胞产生 PGE2的能力与磷脂中 AA 含量呈正相关的关系[40],动物摄食的饲料中各脂肪酸的组成和比例会使磷脂中的 AA 含量发生相应改变,其中的机理如下: ( 1) 磷脂中 AA 的含量与食物中 AA 的摄入量呈正相关关系,鱼体摄入的AA 越多,其免疫细胞中 AA 也相应增加; ( 2 ) 食物中 n-3HUFA ( 如 EPA 和 DHA) 的摄入会竞争AA 加入免疫细 胞 中,导致 AA 的溶入 比 率 下降[41 - 42],从而使 AA 的类花生四烯酸产物 PGE2和 LTB4减少[43 - 45]; ( 3) 食物中富含 18: 2n-6 会导致 AA 的类花生四烯酸产物降低,在鲑上的研究表明,饲喂富含亚油酸或者亚麻酸的植物油会导致包括总红细胞,总白细胞及呼吸爆发在内的非特异性免疫系数的降低,这主要是由于植物油中 ARA 不足,且大量的亚油酸在体内 转 变 成DHGLA ( 20: 3n-6 ) ,DHGLA 作为环氧合酶和脂氧合酶的竞争性底物,会降低 AA 生成类花生四烯酸,结果导致类花生四烯酸的活性成分如 PGE2和 LTB4生 成 量 不 足 以 调 节 正 常 的 免 疫 反应[46 - 49]。Li 等[50 - 51]首次建立了离体大黄鱼巨噬细胞模型,发现随着细胞液中 AA 添加量的增加,前列腺素 PGE2的浓度显著升高,而随着 EPA和 DHA 浓度的升高,PGE2的浓度显著降低,且DHA 较 EPA 具有更强的抑制作用。
总之,导致 PGE2和 LTB4生成量不足的原因有二: 一是 AA 的绝对含量减少; 二是 AA 的相对含量 降 低,这 是 由 于 EPA、DHA 以 及 DHGLA( 20: 3n-6) 竞争抑制环氧合酶和脂氧化酶的底物结合位点,导致 PGE2和 LTB4相应减少。所以食物中 AA 的含量越高,AA /EPA、AA /DHA 以及AA / DHGLA 的比率越高,机体相应产生 PGE2和LTB4的量也越多。
4. 4 细胞因子途径
鱼类免疫系统包括细胞免疫和体液免疫,细胞因子( Cytokine) 是其中重要的组成部分。细胞因子是一类由免疫细胞和非特异性免疫细胞合成或分泌的小分子多肽物质,具有调节多种细胞生理功能的作用。在一般状况下,细胞因子的分泌量很低,或处于失活状态; 但在机体的免疫细胞或组织受到刺激发生新的基因转录后,其含量将会大幅度上升,并识别细胞上高亲和性的表面受体,以协同形式结合其他的细胞因子或者抗病毒分子发挥生物学效应,发挥免疫调节作用[52]。目前已经分离出了包括肿瘤坏死因子( TNF) ,白细胞介素( IL) ,干扰素( IFN) ,一氧化氮合成酶( i NOS)和一些趋化因子( Chemokines) 在内的细胞因子。除此之外,许多实验还证实,鱼类体内细胞因子的结构、作用机制与哺乳动物的具有相似性,但是目前研究较深入的 2 种细胞因子是干扰素( IFN) 和白细胞介素( IL) 。
( 1) 脂肪酸通过干扰素( IFN) 调节免疫
干扰素是脊椎动物体内的一种可溶性蛋白,主要由巨噬细胞分泌,生成速度受温度影响。鱼中的干扰素主要为Ⅰ型包括 IFN-α 和 IFN-β,其基因在多种鱼中被克隆。当生物体受到病毒或者抗原( 如单链 DNA 或者脂多糖) 侵扰后,干扰素可以作用于相应细胞的 IFN-α /β-receptor,然后激活 JAK-STAT 信号转导途径,使相应细胞分泌出一系列具有抗病毒特性的蛋白质,故而在抗病毒的防御机制中发挥着核心调节作用[53]。干扰素调节免疫主要是通过诱导表达抗病毒蛋白来发挥作用,尽管干扰素诱导产生一系列的基因如 Mx、PKR、Viperin、ISG15、IFI56 和 IFI58,但迄今为止,只有鲑和牙鲆的 Mx 蛋白被证实具有抗病毒活性[54]。Montero 等[4]发现不同的脂肪源会影响M x 抗病毒蛋白的表达,M x 抗病毒蛋白是由干扰素控制产生的,所以推测 Mx 在各处理组的表达差异是通过脂肪酸作为转录因子在分子水平上控制了干扰素基因的表达来实现的,其中亚油酸能够提高金头鲷( Sparus aurata) Mx 的表达。
( 2) 脂肪酸通过白细胞介素调节免疫
白介素( IL) 是与鱼类致炎作用以及调理功能密切相关的一个功能性蛋白家族。白介素家族的典型成员包括了 IL-1、IL-2、IL-6 和 IL-8。但目前研究较多也较深入的是 IL-Iβ[52,55 - 56],它主要是由肝脏中被病原激活的巨噬细胞产生,通过结合靶细胞受体 IL-1R 受体复合体,启动早期固有免 疫 反 应,如 刺 激 鲑 和 虹 鳟 ( Oncorhynchusmykiss ) APP ( acute phase proteins ) 的 表 达[56]。Jorgensen 等[57]发现在感染致病菌的鱼中 IL-Iβ的表 达 量 升 高,淋 巴 细 胞 会 相 应 增 殖。Kono等[55]用 DNA 注射方法研究鲤( Cyprinus carpio)对于 IL-Iβ 的免疫反应,发现注射有 IL-Iβ 的鲤淋巴细胞在受到刺激后发生显著增殖,巨嗜细胞吞噬力和超氧阴离子含量也有明显上升,因此对于嗜水气单胞菌( Aeromonas hydrophlia) 的抗性大大增强。AA 的类花生四烯酸产物 LTB4会促使单核细胞和巨噬细胞分泌 IL-1。所以脂肪酸可能是通过活性代谢中间产物来调控 IL 表达,最终调节鱼类免疫的。
4. 5 类固醇激素途径
鱼体在感受到压力后机体的可的松含量会升高,而 可 的 松 水 平 的 上 升 会 引 起 免 疫 抑 制,M ontero 等[58]发现机体在缺乏必需脂肪酸或维生素 E 后机体处于营养不足引发的慢性胁迫中可的松含量会显著上升,从而抑制鱼体免疫性能的发挥。同样,在金头鲷中用亚麻酸取代 80% 的鱼油实验结束后进行拥挤压力实验,结果发现,亚麻油组鱼体的可的松水平明显高于鱼油组。这是由于脂肪酸缺乏或者比例失调会刺激丘脑 - 垂体 -肾上腺轴( hypothalamic-pituitary-interrenal,HPI) ,使得血浆中促肾上腺皮质激素和肾上腺皮质激素浓度上升,从而抑制机体免疫力。脂肪酸调节HPI 来控制可的松含量的机理虽还不清楚,但有些学者将其也作为一个免疫指标[2,58]。
5 展望
脂肪酸与鱼类的免疫性能具有高度相关性,而调控机制的研究尚有较大空间。未来的研究应该侧重于以下几个方面: ( 1) 脂肪酸是如何通过转录因子调节鱼类免疫的; ( 2) 脂肪酸是如何通过改变肠道脂肪酸组成或菌群结构调控鱼类免疫的; ( 3) 脂肪酸介导的免疫因子与鱼体脂肪代谢和免疫力的关系; ( 4) 非脂肪酸成分( 矿物质、维生素) 调控鱼类脂肪酸代谢和免疫过程的机制。
